Key Findings
A study led by Professor Motoshi Suzuki and graduate student Dat Quoc Tran at Fujita Health University, Japan, investigated whether inherited genetic factors contribute to multiple primary lung cancers.
Researchers analyzed 26 lung adenocarcinoma (LUAD) tumors from 11 patients and screened 125 tumors from an independent cohort of 123 patients. They identified two novel truncating germline variants in BRCA2 in two unrelated patients, suggesting inherited susceptibility to multiple primary lung cancers. Genomic profiling helped distinguish multiple primary tumors from intrapulmonary metastases in 9 out of 11 patients. The findings may support future use of PARP inhibitors for patients with these genetic backgrounds.
Background
Lung cancer is the leading cause of cancer death worldwide. At Fujita Health University Hospital, 7.5% of lung cancer surgeries involved patients with multiple lung lesions, raising questions about whether these represent metastasis or independent primary cancers linked to inherited predisposition.
Implications
Molecular profiling can improve diagnostic accuracy for multiple lung lesions. Identifying inherited BRCA2 alterations opens possibilities for targeted therapies similar to those used in other BRCA-associated cancers.
